In mice, SLU-PP-332 moves fast. Gene activity shifts within one to three hours, fuel use swings toward fat inside about two hours, and body composition changes over roughly 28 days. In people, how long SLU-PP-332 takes to work has never been measured. No published human trial, no human dosing data, no approval from any regulator anywhere.
That gap is the answer. Most pages ranking for this question bury it.
I run the regenerative side of Formation, a regenerative medicine clinic in Tampa. We don’t offer this compound and we can’t. It’s a research-grade molecule. So this piece covers what the animal data shows, why those hours and weeks don’t transfer to your body, and what we reach for instead. What it won’t give you is a dose or a protocol.
SLU-PP-332 is a synthetic small molecule that switches on three estrogen-related receptors: ERR alpha, ERR beta, and ERR gamma. Researchers at Saint Louis University built it to copy the gene activity of endurance exercise without the exercise. It isn’t a peptide. Every published result comes from cell cultures and mice.
The name throws people. Estrogen-related receptors have nothing to do with estrogen. They govern how cells build and spend energy.
Patients ask me about this molecule constantly, which tells you how far the conversation has drifted from the lab bench where SLU-PP-332 is actually used.
Fast, if you’re a mouse. The published windows sort into three tiers.

The endurance number made this compound famous. In the 2023 ACS Chemical Biology work, one dose given an hour before treadmill testing raised running distance by roughly 45 percent and running time by roughly 70 percent against control animals.
Read the fine print. Those were untrained mice, and untrained animals have the most room to improve. The dose went in as an injection to the abdominal cavity, which nobody does outside a lab. And the authors themselves describe what they built as a chemical tool for use in animals, not a finished drug.
The earliest measurable change is a gene called DDIT4 turning on, detectable at one hour and settling back to baseline by six.
DDIT4 works like a switch that reorganizes how a cell spends energy. Exercise flips the same switch. That overlap is the real finding here, and it’s a good one.
Metabolism moves almost as fast. Researchers track something called respiratory exchange ratio, a breath measurement telling you whether an animal is burning mostly sugar or mostly fat. In the 2024 Journal of Pharmacology and Experimental Therapeutics paper, that ratio dropped within two hours of the first dose and calculated fat oxidation rose about 25 percent against controls. The animals were diet-induced obese or genetically obese mice, so their starting metabolism was already impaired.
Structural change needs about two weeks. Muscle fiber composition and grip strength both shifted measurably around day 13 of continuous dosing.
Body composition takes a month. Over 28 days, diet-induced obese mice finished roughly 12 percent lower in body weight with far less fat gain, plus better glucose handling and less liver fat. Food intake didn’t consistently change, which separates this pathway from appetite suppression and from most every peptide for fat loss people ask me about.
One detail nobody quotes. The genetically obese arm ran about 12 days instead of 28, and the published reason was tolerability of twice-daily abdominal injections. When the dosing schedule itself is hard on the animals, that tells you something about the schedule.
Twenty-eight days is also the ceiling for the standard protocol. A separate kidney-aging model ran eight weeks. Past that, published data stops.
Three reasons, and any one of them is enough.
Route. Every efficacy study used injection straight into the abdominal cavity. Oral absorption of the parent compound is poor. It barely dissolves, and in human liver preparations it clears with a half-life around 31 minutes. A molecule that disappears that fast in a human liver test is a chemistry problem waiting on a better version, which is exactly why successor compounds exist.
Scaling. Published mouse doses ran 25 to 50 mg/kg. Converting that to a person takes surface-area math plus real pharmacokinetic modeling, and none of it has been validated for this molecule.
Measurement. Nothing has been measured in a human being. Not absorption, not duration, not response.
Every human timeline circulating online is a mouse study wearing a costume, which is why the benefits of SLU-PP-332 stay unproven in people.
Competitor pages skip this part. The heart question isn’t closed.
Pan-ERR agonists were tested in a mouse heart-failure model, published in Circulation in 2024. They improved pumping function and reduced scarring, which reads as good news. The same work showed these compounds did not affect cardiac hypertrophy, meaning they didn’t stop the heart muscle from thickening under stress. ERR gamma sits in heart tissue, and its activation has been tied to hypertrophy markers in other preclinical models.
Mind the limits. A surgical pressure-overload model, six weeks, in mice.
Nobody is claiming harm in humans. Nobody can rule it out either, and a compound with an open cardiac question and zero human safety data is not one I’d hand to someone chasing a leaner midsection.
No. There’s no legal route to a person: no prescription pathway, no compounding pathway, no trial to enroll in.
The research-use-only label doesn’t change what something is. The FDA has warned companies selling unapproved drugs falsely labeled for research purposes or not for human consumption, noting those products went directly to consumers with dosing instructions attached. The disclaimer is decoration.
For us that settles it. Administering, prescribing, purchasing, or dispensing a research-grade molecule is illegal. Formation holds SSRP and ISSCA certification, the bodies that certify peptides, exosomes, and stem cells. Those certifications carry real sourcing requirements, and they’d mean nothing if we made exceptions.
Human-grade compounds, chosen for a specific pathway, layered onto a lifestyle already doing the heavy lifting.
Human-grade only isn’t a house preference. States have started writing it into law, and research-grade peptides drew an outright ban in Alabama.
Dr. Grandhige built this practice around being willing to say no, and that shows up more in regenerative medicine than anywhere else in the building. Sometimes the honest answer is that the molecule isn’t ready.
Think of your physiology as a seesaw. One side builds: protein, resistance training, growth signaling, repair. The other side renews through fasting windows, cellular cleanup, and clearing out damaged parts. Health lives in the movement between them, not parked at either end.
Nature already runs this way. Day and night. Feeding and fasting. Training and recovery.
An exercise mimetic pushes the renewal side hard and holds it there. Most trouble I see comes from people camped on one side permanently, always eating and never fasting, always training and never recovering.
There’s a second problem with leaning on one molecule. Push a single pathway hard enough and the body answers through a different one. Drive fat oxidation while sleep is broken and stress hormones run high, and the body claws the difference back somewhere you weren’t watching. So we come at a goal from several directions at once.
The lifestyle work does the work. Zone 2 cardio, lifting three or four days a week, protein at every meal, a real overnight fast, sleep you actually protect. Compounds sharpen what’s already moving. They don’t move it for you.
Hallmarks addressed: mitochondrial dysfunction and deregulated nutrient-sensing.
ERR activation raises mitochondrial building and fat oxidation, which speaks to mitochondrial dysfunction, where energy production falls and oxidative stress climbs. The fuel-switching effect speaks to deregulated nutrient-sensing, the loss of flexibility between burning sugar and burning fat.
Both hallmarks have human-grade routes. SS-31 peptide targets mitochondrial membranes directly. Metabolic flexibility responds to fasting structure and zone 2 work faster than most people expect. Ask three questions and the noise clears quickly.
Is there human data, meaning an actual person in a published study rather than a mechanism or a mouse? What route did the study use, since an injection into a mouse abdomen tells you nothing about a capsule? How long did the longest study run?
Run SLU-PP-332 through that filter and you get: none, intraperitoneal, 28 days. Run the most popular peptides through the same filter and you get a very different picture.
